The cell cultures of both plants were established separately. In this study some basic experiments were also done due to the fact that no research reports were found on C. carvi in case of seed germination, callus induction and establishment of cell suspension cultures. The cells of B. persicum and C. carvi were combined together in different ratios (100:0, 25:75, 50:50 and 75:25) to study their interaction on cumin aldehyde production in a co-culture system. For Gas chromatography (GC) analysis of cumin aldehyde, the cells and medium were homogenized and then extracted with ethyle acetate at room temperature and then analyzed by GC. The results revealed that in contrast to black zira for seed germination process, Caravie seeds do not need chilling. It has also been found that sucrose at the level of 3% is suitable for increasing the biomass of Caravie cells in MS medium. The results of co-culture of B. persicum and C. carvi cells demonstrated that at the combination of B. persicum (25%) and C. carvi (75%) the production of cumin aldehyde could be increased to the level of 23.1 mg/100 g extract after five weeks which is higher than B. persicum (100%) at similar period with 17.5 mg/100 g extract. It is also found that the production of cumin aldehyde has an increasing trend till 5th week and then decreased to the minimum level in the 7th week.}, keywords = {Bunium Persicum,Carum carvi,cell suspension culture,cumin aldehyde,co-culture}, title_fa = {بررسی تولید کومین آلدئید به‌عنوان طعم‌دهندۀ مواد غذایی با استفاده از سیستم کشت توأم سلولی زیرۀ سیاه کوهی Bunium persicum [Bioss] و زیرۀ سیاه اروپایی .Carum carvi L}, abstract_fa = {در این پژوهش به‌منظور افزایش تولید کومین آلدئید به روش کشت سلولی تعلیقی از سیستم کشت توأم سلولی دو گیاه زیرۀ سیاه کوهی  [Boiss] Bunium persicum  و زیرۀ سیاه اروپایی  L. Carum carvi استفاده شد. براین‌اساس بذور دو گیاه در یک محیط کشت ساده و در شرایط دمایی 4 و 25 درجه سانتی‌گراد جوانه‌زنی انجام دادند. جهت القای کالوس از بذر جوانه زدۀ دو گیاه در محیط کشت جامد MS استفاده شد. سپس برای رسیدن به حداکثر بیوماس سلولی در محیط کشت جامد MS، غلظت‌های مختلف ساکارز 6-1 درصد موردِبررسی قرار گرفت. نتایج نشان داد که غلظت 3 درصد ساکارز پس از 2 هفته به حداکثر وزن تر سلول به میزان 0/21 گرم می‌رسد. پس از استقرار کشت تعلیقی سلولی هر کدام از گیاهان به­طور جداگانه، سلول‌های حاصل از کشت تعلیقی دو گیاه زیرۀ سیاه کوهی و زیرۀ سیاه اروپایی به نسبت درصدهای مختلف (75:25، 50:50، 75:25، 100:0) با یکدیگر مخلوط شده و روی شیکر در شرایط محیطی همانند مرحلۀ کشت کالوس قرار گرفتند. اندازه‌گیری کومین آلدئید به روش کروماتوگرافی گازی از عصارۀ حاصله  از نمونه‌های کشت توأم سلولی  طی 7 هفته مشخص نمود که بیشترین میزان تولید کومین آلدئید در تمام نسبت درصدها مربوط به ترکیب 25 درصد زیرۀ سیاه کوهی و 75 درصد زیرۀ سیاه اروپایی به میزان 23/1 میلی‌گرم در 100 گرم در هفته پنجم می‌باشد که در مقایسه با زیرۀ سیاه کوهی که 7/61 میلی‌گرم در 100 گرم بود در همین مدت سه برابر افزایش تولید را نشان می‎دهد. همچنین کمترین میزان کومین آلدئید در این ترکیب مربوط به هفته اول بود.}, keywords_fa = {زیرۀ سیاه کوهی,زیرۀ سیاه اروپایی,کشت توأم سلولی,کشت سوسپانسیون سلولی,کومین آلدئید}, url = {https://journals.rifst.ac.ir/article_68135.html}, eprint = {https://journals.rifst.ac.ir/article_68135_29f3ee41fe52c19d27b39adfca192e05.pdf} }
