Monascus pigments usually accompanied with citrinin contamination which limits their wide application in foods. The usage of Saccharomyces cerevisiae is known as a useful biological way for citrinin elimination. In this study, the effect of  Saccharomyces cerevisiae cells in twomodes of treatment (heat treatment at 121  °C and acid treatment in 2M HCL) and live at three different concentrations (103, 104, 105 CFU/mL) were investigated on the amount of citrinin and yellow, orange and red pigments produced by Monascus purpureus. The results showed that live yeast cell and yeast cells treated by heat and acid significantly decreased the citrinin in culture, so that citrinin content decreased from 4.43 mg/L in control sample to 0.9, 0.10 and 0.07 mg/L (at 105 CFU/mL yeast suspension) respectively. The maximum amount of yellow, orange and red extracellular and intracellular measured pigments belong to the samples with live yeast, while the amount of this pigments in the samples containing yeast treated with heat and acid reduced respectively. Among the treatments, the use of live yeast at concentration of 104 CFU/mL per milliliter provided the best conditions to achieve the desired goal.}, keywords = {Acid treatment,Heat treatment,pigment,Saccharomyces cerevisiae Citrinin,Monascus purpureus}, title_fa = {بررسی اثر مخمر ساکارومایسس سرویزیۀ زنده و تیمارشده با حرارت و اسید بر میزان سیترینین و رنگدانه‌های موناسکوس پورپورئوس}, abstract_fa = {کپک موناسکوس پورپورئوس به‌دلیل تولید دامنۀ گسترده‌ای از رنگدانه‌های طبیعی، توجه بسیاری از تولیدکنندگان مواد غذایی را به‌عنوان جایگزینی برای رنگ‌های سنتزی جلب کرده است. رنگدانه‌های موناسکوس معمولاً آلوده به سیترینین هستند که همین امر کاربرد گستردۀ آنها را در مواد غذایی محدود می‌کند. استفاده از مخمر ساکارومایسس سرویزیه یک روش بیولوژیکی مناسب برای حذف توکسین شناخته می‌شود. در این پژوهش اثر مخمر ساکارومایسس سرویزیه در دو حالت غیرفعال‌شده (تیمار حرارتی در دمای 121 درجۀ سانتی‌گراد و تیمار اسیدی در اسید هیدروکلریک دو مولار) و زنده در سه غلظت مختلف (103، 104 و 105 واحد تشکیل کلنی در میلی‌لیتر) بر میزان سیترینین و رنگدانۀ زرد، نارنجی و قرمز تولیدشده توسط کپک موناسکوس پورپورئوس موردبررسی قرار گرفت. نتایج به‌دست‌آمده نشان داد که مخمر زنده و تیمارشده با حرارت و اسید به‌طور قابل‌توجهی باعث کاهش سیترینین در محیط شد، به‌طوری‌که میزان سیترینین از 43/4 میلی‌گرم بر لیتر در نمونۀ کنترل فاقد مخمر به‌ترتیب به 0/9، 0/1 و 0/07 میلی‌گرم بر لیتر (در غلظت 105 واحد تشکیل کلنی در میلی‌لیتر از سوسپانسیون مخمری) کاهش یافت. حداکثر مقدار رنگدانه‌های خارج سلولی و داخل سلولی زرد، نارنجی و قرمز اندازه‌گیری‌شده متعلق به نمونه‌های حاوی مخمر زنده بود، درحالی‌که مقدار این رنگدانه‌ها در نمونه‌های حاوی مخمر تیمارشده با حرارت و اسید به‌ترتیب کاهش داشتند. از بین تیمارها، استفاده از مخمر زنده در غلظت 104 واحد تشکیل کلنی در میلی‌لیتر بهترین شرایط را برای رسیدن به هدف موردنظر فراهم کرد.}, keywords_fa = {تیمار اسیدی,تیمار حرارتی,رنگدانه,ساکارومایسس سرویزیۀ سیترینین,موناسکوس پورپورئوس}, url = {https://journals.rifst.ac.ir/article_109383.html}, eprint = {https://journals.rifst.ac.ir/article_109383_820aa23ad6f511f82367347823129927.pdf} }
