بازدهی استخراج بتا-دی-گلوکان از ضایعات قارچ دکمهای (آگاریکوس بیسپوروس) و توانایی آن در جذب آفلاتوکسین B1
بتاگلوکانها در گسترۀ وسیعی از موجودات مختلف ازجمله مخمر، قارچ، باکتری، جلبک، جو و جو دوسر یافت میشود و اثرات بیولوژیکی متنوعی را از خود نشان میدهند. بتاگلوکانها از پیوند بتاگلوکوزیدی واحدهای D-گلوکز به یکدیگر تشکیل شدهاند. توانایی جذب سموم قارچی ازجمله آفلاتوکسینها توسط بتاگلوکان در سالهای اخیر بسیار موردتوجه قرار گرفته است. آفلاتوکسینها گروهی از ترکیبات سمی با قدرت سرطانزایی بالا هستند که بهعنوان یک عامل آلودهکنندۀ مواد غذایی برای انسان و حیوان، شناخته میشوند. آفلاتوکسین B1 سمیترین ترکیب در ایجاد سرطان در بین انواع آفلاتوکسینهاست. توانایی جذب سموم قارچی توسط بتاگلوکان بهطور مستقیم به عواملی مانند ساختمان مولکولی، روش استخراج و منبع حاوی بتاگلوکان بستگی دارد. بتاگلوکان مستخرج از منبع قارچی دارای اتصالات بتا (3-1) در رشتۀ اصلی و بتا (6-1) در رشتۀ جانبی و در محل انشعابهاست. وجود انشعابهای زیاد با طول کوتاه با ساختاری شانهمانند، سبب افزایش جذب سموم قارچی توسط این ترکیب میشود. در تحقیق حاضر، برای اولین بار میزان بازدهی چندین روش استخراج بتاگلوکان از دیوارۀ سلولی ساقههای دورریز قارچ دکمهای (آگاریکوس بیسپوروس) اندازهگیری و توانایی جذب آفلاتوکسین B1 نیز سنجیده شد. نتایج نشان داد علیرغم اینکه میزان بازدهی استخراج بتاگلوکان به روش اسیدی بیشتر از سایر روشهاست (20/5 درصد)، اما براساس سنجش به روش HPLC، بتاگلوکان استخراجشده با استفاده از قلیای داغ، توانایی جذب و خارجکردن 90/2 درصد آفلاتوکسین B1 را از نمونۀ آلودهشده را دارد

تعیین شرایط بهینۀ آبگیری اسمزی بهعنوان پیشتیمار در خشککردن با هوای داغ برشهای شلغم به روش سطح
هدف از مطالعۀ حاضر، بررسی اثر غلظت محلول اسمزی (30، 45 و 60 درصد وزنی/وزنی)، دمای محلول اسمزی (30، 40 و 50 درجۀ سانتیگراد) و زمان غوطهوری (4، 5 و 6 ساعت)، بر خروج آب (WL)، جذب مادۀ جامد (SG)، کاهش وزن (WR)، محتوی ویتامین C، چروکیدگی، نسبت بازآبپوشی (RR) و شاخصهای رنگ (L، a و b) طی آبگیری اسمزی- خشککردن ورقههای شلغم بود. روش سطح پاسخ (RSM) نیز برای یافتن شرایط بهینه مورداستفاده قرار گرفت. نتایج نشان داد که طی آبگیری اسمزی نمونههای شلغم، متغیرهای دمای محلول اسمزی، غلظت محلول و زمان غوطهوری تأثیر معنیداری بر پارامترهای انتقال جرم (WL، SG و WR)، محتوی ویتامین C، چروکیدگی، RR و شاخص رنگ (L) داشتند. شرایط بهینۀ آبگیری اسمزی برای شلغم، دمای محلول اسمزی 30/81 درجۀ سانتیگراد، غلظت محلول اسمزی 60 درصد و زمان غوطهوری 6 ساعت بود. تحت این شرایط، مقادیر پاسخهای WL، SG، WR، چروکیدگی، نسبت بازآبپوشی (RR)، محتوی ویتامین C و شاخصهای رنگی L، a و b بهترتیب 83/10، 12/91 و 70/19 درصد، 27/76، 4/19 و 11/64 (میلیگرم در 100 گرم مادۀ خشک) و 33/85، 25/49 و 15/91 بود. نتایج پژوهش حاضر میتواند جهت فراوری حداقلی برشهای شلغم با استفاده از آبگیری اسمزی و خشککردن بُعدی نمونهها مورداستفاده قرار گیرد.

غلتکهای نرمکننده در فرایند تولید آرد نقش مهمی دارند و ازآنجاییکه افزایش کیفیت آرد مورداستفاده در صنایع پخت بهعنوان مهمترین فاکتور در بهبود خصوصیات کیفی و تغذیهای نان بشمار میآید، هدف از انجام این پژوهش ارزیابی تأثیر حذف غلتکهای نرمکنندۀ C1A، C1B، C2، C3 و C5 بر ویژگیهای فیزیکوشیمیایی آرد و خصوصیات رئولوژیکی خمیر و انتخاب مؤثرترین غلتک در فرایند آسیابانی در یک طرح کاملاً تصادفی بود (05/0≥p ). نتایج نشان داد که دو نمونۀ آرد که در فرایند آسیابانی آنها غلتکهای C5 و C3 حذف شده بودند، از میزان رطوبت، خاکستر، اندازۀ ذرات کوچکتر از 125 میکرون و نشاستۀ آسیبدیدۀ کمتری برخوردار میباشند. همچنین نمونهای که در فرایند آسیابانی آن غلتک C3 حذف شده بود از کمترین میزان گلوتن (25 درصد)، اندیس گلوتن (70)، حجم رسوب زلنی (17 میلیمتر) برخوردار بود. همچنین با حذف غلتک C3 جذب آب آرد، زمان توسعه، زمان پایداری و ارزش والوریمتری بیشترین کاهش را نسبت به نمونۀ شاهد نشان داد. بنابراین مشخص شد آردی که در فرایند آسیابانی آن، غلتک C3 حذف شده است، ویژگیهای فیزیکوشیمیایی آرد و خمیر به شدت تضعیف گردید. در ادامه این پژوهش با آرد حاصل از این نمونه و نمونۀ شاهد نان بربری تولید شد و نتایج نشان داد که میزان سفتی بافت نمونه با حذف غلتک C3 نسبت به نمونۀ شاهد در هر دو بازۀ زمانی (2 و 72 ساعت پس از پخت) بیش از 35 درصد افزایش یافت. همچنین در بخش ارزیابی حسی، نمونۀ شاهد امتیاز پذیرشکلی بیشتری را به خود اختصاص داد. بنابراین باتوجهبه نتایج این تحقیق مشخص شد که حذف غلتک C3 نسبت به سایر غلتکهای نرمکننده بیشترین تأثیر را بر کیفیت آرد، خمیر و محصول نهایی دارد.

تأثیر بستهبندی با اتمسفر اصلاحشده و نوع مادۀ بستهبندی بر ماندگاری مغز گردو
گردو با داشتن اسیدهای چرب ضروری و غیراشباع گروه امگا-3، بهعنوان یکی از مهمترین خشکبار ازنظر ارزش غذایی مطرح است. ژنوتیپهای 25 و 29 (ژنوتیپهای برتر گردو) با هدف بررسی تأثیر بستهبندی به روش اتمسفر اصلاحشده بر ترکیبات سودمند موجود در روغن گردوی آنها، انتخاب شدند. مغز گردوها تحت شرایط اتمسفر اصلاحشده داخل پلاستیک پنجلایه متالیزه با ضخامت 90 میکرون بستهبندی گردیدند. بعد از استخراج روغن به روش سرد و خالصسازی آن، ترکیب کمی و کیفی اسیدهای چرب تشکیلدهنده اندازهگیری شد. میزان اسیدهای چرب، مقدار آفلاتوکسین، عدد پراکسید، عدد یدی، اسیدیته و ارزیابی حسی نیز در روز صفر و طی یکسال نگهداری تعیین گردید. نتایج نشان داد میزان اسیدهای چرب غیراشباع نسبت به اسیدهای چرب اشباع بیشتر بوده و اسید لینولئیک، اسید چرب غالب است. مقدار اسیدهای چرب اشباع نیز کمتر از 10 درصد تعیین شد. میزان آفلاتوکسین در ژنوتیپ 25 و 29، بهترتیب صفر و 5 قسمت در بیلیون برآورد گردید. میزان تغییرات رطوبت و اندیس پراکسید نمونههای مغز گردو طی یکسال نگهداری، بهترتیب حدود 4/32-1/55 درصد و 4/65- 0/48 میلیاکیوالان اکسیژن بر کیلوگرم متغیر بود. بیشترین میزان اسیدیته در مغز گردوی رقم 25 مشاهده شد. با افزایش زمان نگهداری، عدد پراکسید، اسیدیتۀ قابل تیتر و اُفت وزن افزایش، و عدد یدی کاهش یافت. بهطورکلی، ژنوتیپ 25 مغز گردوی بستهبندیشده در فیلم پنجلایه با اتمسفر اصلاحشده حاوی 6-5 درصد اکسیژن، 15 درصد دیاکسیدکربن و 80-79 درصد ازت، بهدلیل نداشتن آفلاتوکسین و خواص شیمیایی و حسی مطلوب، قابل توصیه است.

بررسی اثر اضافهکردن اسانس پوست پرتقال بر ویژگیهای شیمیایی، میکروبی و حسی لواشک آلو
امروزه با افزایش آگاهی مصرفکنندگان از ایمنی و کیفیت مواد غذایی، تقاضا برای استفاده از مواد غذایی بدون نگهدارنده (مصنوعی) و حاوی افزودنیهای طبیعی افزایش یافته است. گیاهان یکی از منابع اصلی ترکیبات ضدمیکروبی و حاوی اسانسهای روغنی میباشند که دارای تأثیرات محافظتکنندگی در برابر میکروارگانیسمها هستند. در این پژوهش اثر اضافهکردن اسانس پوست پرتقال بر ویژگیهای شیمیایی، میکروبی و حسی لواشک آلو طی دورۀ 15 روزه موردبررسی قرار گرفت. نتایج نشان داد که افزودن اسانس پوست پرتقال بهطور معنیداری (0/05>p ) باعث کاهش استحکام کششی و افزایش مقاومت به کشش لواشک آلو گردید. در انتهای دورۀ نگهداری 15 روزه لواشک آلو بالاترین مقدار pH و پایینترین مقدار اسیدیته در نمونۀ حاوی 0/5 درصد اسانس پوست پرتقال مشاهده گردید. در هیچ کدام از نمونههای لواشک آلو حاوی اسانس پوست پرتقال کپک و مخمر مشاهده نگردید. نمونۀ حاوی 0/5 درصد اسانس پوست پرتقال دارای بالاترین میزان باکتری اسیدلاکتیک و نمونۀ حاوی 0/1 درصد اسانس پوست پرتقال کمترین میزان باکتریهای اسیدلاکتیک را نشان دادند. نتایج حاصل از این پژوهش نشان داد که نمونۀ لواشک حاوی 0/5 درصد اسانس پوست پرتقال کمترین و نمونۀ شاهد بیشترین باکتریهای کلی را دارا بودند. نتایج تجزیۀ واریانس، اختلاف معنیداری را در خاکستر لواشکها نشان نداد. بیشترین امتیاز بافت، مزه و آروما مربوط به نمونۀ شاهد بود. پذیرش کلی نمونۀ شاهد و نمونۀ پوست پرتقال 0/1 درصد طعم آلو بیشترین بود. بهترتیب در این دو نمونه طعم پرتقال بیشتر حس میشد. باتوجهبه خاصیت ضدمیکروبی اسانس پوست پرتقال و تأثیرات مثبت آن در ویژگیهای بافتی میتوان از آن بهعنوان یک نگهدارندۀ طبیعی در فراوردۀ لواشک آلو استفاده کرد.

پیشبینی تغییرات کیفی کپور نقرهای بستهبندیشده در شرایط خلأ با کمک روش سطح پاسخ
هدف از مطالعۀ حاضر استفاده از مدلسازی مرکب برای پیشبینی تغییرات میکروبی، میزان بازهای نیتروژنی فرار (TVB-N)، آنالیز حسی و نیز فاکتور تازگی فیلۀ کپور نقرهای (Hypophthalmichthys molitrix) بستهبندیشده در شرایط خلأ بود. ماهیان بستهبندیشده در خلأ در دماهای صفر، 6 و 12 درجۀ سانتیگراد که براساس خروجی نرمافزار State-Ease Design Expert بهینهشده بودند، به مدت 5، 10 و 15 روز نگهداری شدند. درخصوص پارامترهای کیفی فیلۀ کپور نقرهای بستهبندیشده، بین تمامی تیمارها در زمان و در درجهحرارتهای موردبررسی اختلاف معنیداری مشاهده شد (0/05>p ). دو پارامتر اینوزینمونوفسفات (IMP) و اینوزین (HxR) با افزایش زمان و دما روند کاهشی را نشان دادند (0/05>p ). شاخص تازگی و آنالیز حسی در مدت زمان کوتاه (5 روز) در حالت مجاز و بهینه قرار داشتند. در این آزمون مقادیر پیشبینیشده با درصد مقبولیت 78 درصد با مقادیر برآوردهشده همخوانی داشتند. مشاهدات بیانگر همبستگی خوب بین نتایج بهدستآمده با روش تجربی و مقادیر پیشبینیشده با روش آماری بود. مدلهای موردبررسی از ضریب تبیین (R2) و ضریب تبیین اصلاحشده (R2-adj) نزدیک به 1 برخوردار بودند، که نشان میدهد مدل آزمایشی بهکارگرفتهشده، قادر به بررسی پیشبینی تغییرات کیفی فیله با درصد خطای پایین بود. موفقیت این کار میتواند سبب بهبود مدلهای جنبشی در صنعت غذا شود، لذا کاربرد آن به ماندگاری محصولات غذایی کمک شایانی میکند.

ارزیابی فساد شیمیایی و میکروبی فیلۀ مرغ پوششدادهشده با کیتوزان، اسانس زنجبیل و کنسانترۀ ازگیل، طی نگهداری در دمای یخچال
اکسیداسیون لیپیدها و رشد میکروبی ازجمله عوامل مؤثر بر خصوصیات کیفی و فسادپذیری گوشت در طی نگهداری است. در این مطالعه بهکارگیری پوشش خوراکی کیتوزان، کنسانترۀ ازگیل، اسانس زنجبیل به تنهایی و در ترکیب با یکدیگر برکاهش فساد شیمیایی و افزایش زمان ماندگاری گوشت مرغ نگهداریشده در دمای یخچال موردبررسی قرارگرفت. 8 گروه موردبررسی طی یک دورۀ 12 روزه در دمای یخچال نگهداریشده و ازنظر شاخصهای شیمیایی اکسیداسیون اولیه (PV) ، ثانویه (TBA) و شاخص بازهای فرار (TVB-N) و میکروبیولوژیکی (شمارش باکتریهای مزوفیل هوازی و سرمادوست)، ارزیابی حسی در روزهای صفر، 4، 8 و 12 موردبررسی قرار گرفتند. تست توتال فنل و قدرت احیاکنندگی نیز بهمنظور بررسی خواص ضداکسایشی انجام گردید. براساس نتایج حاصل از دستگاه گاز کروماتوگراف-طیفسنج جرمی (GC-MS) ترکیبات اسانس زنجبیل آلفا-زینجیبرن (36/54 درصد)، بتا-سسکوایفلاندرن (16/45 درصد) و ترانس-گاما-کادینن (10/27 درصد) بودند. نتایج نشان داد که در تیمارهای پوششدادهشده با کیتوزان حاوی اسانس زنجبیل 2 درصد و کنسانترۀ ازگیل شمارش شاخصهای میکروبی ذکرشده در مقایسه با گروه کنترل بهصورت معنیداری کاهش یافت (0/05>p ) و شاخصهای اکسیداسیون در مدت زمان نگهداری، تغییرات کمتری داشته و با اختلاف معنیدار (0/05>p )، قویترین اثر ضداکسایشی و حسی نسبت به سایر گروهها را به خود اختصاص داد. نتایج میکروبیولوژیکی، شیمیایی و حسی این مطالعه نشان داد که اثر پوشش کیتوزان همراه با کنسانترۀ ازگیل و اسانس 2 درصد باعث حفظ کیفیت و افزایش ماندگاری به مدت 12 روز در در شرایط یخچالی شد.

توسعۀ کاربرد پوشش خوراکی جدید حاوی صمغ شاهی و مقایسۀ اثرات آن با رومالهای سنتی نان بدون گلوتن بر پایۀ آرد سورگوم
رومالها جهت بهبود بافت و ویژگیهای ظاهری نان استفاده میشوند. در این پژوهش اثرات رومالهای سنتی (آب، روغن، پودر پنیر و صمغ زانتان) و رومال حاوی صمغ شاهی بر ویژگیهای فیزیکوشیمیایی و خصوصیات حسی نان بدون گلوتن مقایسه شدند. بدینمنظور حجم مخصوص، تخلخل، فعالیت آبی، رطوبت، سفتی بافت و ویژگیهای حسی نان بررسی گردید. نتایج نشان داد، نمونههای حاوی پودر پنیر و روغن کمترین میزان رطوبت و نمونههای حاوی صمغ شاهی و زانتان بیشترین حجم مخصوص و تخلخل را داشتند. همچنین نتایج حاکی از آن بود نمونههای تیمارشده با رومال آب و صمغ شاهی دارای کمترین میزان بیاتی و سفتی بافت بودند. این در حالی بود که تمام رومالهای استفادهشده در این پژوهش اثر معنیداری (0/05<p ) بر بو و مزۀ نمونههای تولیدی نداشتند. درنهایت باید گفت، صمغ شاهی بهعنوان یک رومال جدید عملکرد بهتری در مقایسه با صمغ زانتان داشت و از رومال حاوی این صمغ میتوان جهت بهبود سطح نو ویژگیهای کیفی و ظاهری نان بدون گلوتن استفاده نمود.
