اثر پودر آبپنیر و بیکربنات سدیم بر مقدار آکریلآمید در بیسکویتهای پتیبور و مادر
آکریلآمید ترکیبی سرطانزاست که مواد غذایی غنی از کربوهیدرات مانند بیسکویت مستعد تشکیل آن میباشند. از آنجایی که بیسکویت یکی از پرمصرفترین فرآوردههای آردی به عنوان میانوعده بهویژه در بین کودکان است؛ در پژوهش حاضر اثر جایگزینی شیرخشک با پودر آب پنیر در سه سطح (25، 75 و 100 درصد) و اثر جایگزینی بیکربنات سدیم با بیکربنات آمونیوم و بکینگپودر بر میزان آکریلآمید، قند احیاء، پروتئین، رطوبت، pH و اسید قابل تیتر در دو نوع بیسکویت پتیبور و مادر بررسی شد. با افزایش درصد جایگزینی پودر آب پنیر در دو نوع بیسکویت پتیبور و مادر،pH تنها در نمونههای بیسکویت تهیه شده از شیرخشک کامل و پودر آب پنیر کامل با یکدیگر تفاوت معنیدار نشان داد. همچنین میزان پروتئین روند کاهشی و اسید قابل تیتر و قند احیاء روند افزایشی نشان دادند. میزان آکریلآمید در سطح جایگزینی 100 درصد پودر آب پنیر نسبت به نمونه شاهد در بیسکویت مادر و پتیبور به ترتیب 11/9 و 4/1 درصد کاهش یافت. با جایگزینی بیکربنات سدیم با بکینگپودر در فرمول نمونههای بیسکویت، pH کاهش و اسید قابل تیتر افزایش یافت. در صورت استفاده از بیکربنات سدیم در فرمول بیسکویتهای پتیبور و مادر، محتوی قند احیاء بهطور معنیداری کمتر بود. کمترین و بیشترین میزان آکریلآمید به ترتیب در نمونههای بیسکویت حاوی بیکربنات سدیم و بیکربنات آمونیوم مشاهده شد.

ارزیابی تخلیص آفلاتوکسینها از نمونههای فلفل با استفاده از ستونهای مولتی فانکشنال و مقایسه آن با روش ستونهای ایمونوافینیتی
هدف از انجام این مطالعه مقایسه دو روش خالصسازی با استفاده از ستونهای ایمینیوافینیتی و مولتی فانکشنال (مایکوسپ)، برای اندازهگیری آفلاتوکسینهای B1، B2، G1 و G2 توسط کروماتوگرافی مایع با کارآیی بالا (HPLC) در فلفل سیاه، فلفل سفید و فلفل قرمز بود.نمونههای فلفل سیاه، سفید و قرمز عاری از سم تهیه و با مقادیر 1، 5 و 10 نانوگرم بر گرم از هریک از سموم آلوده شدند. خالصسازی با استفاده از هر دو ستون ایمونوافینیتی و مایکوسپ انجام و مقدار بازیابی سم به هر دو روش اندازهگیری شد. شش نمونه فلفل نیز از بازار تهیه و آفلاتوکسینها در هر 6 نمونه به هر دو روش خالصسازی و اندازهگیری شدند. دادهها با استفاده از آنالیز واریانس تجزیه و تحلیل شد. بازیابی سموم با استفاده از ستون ایمونوافینیتی در محدوده 6/6 ±69/8 برای سم G2 و 5/7±101/5 برای سم B1 بود. برای ستون مایکوسپ نیز مقادیر بازیابی بین 3/4±70/9 برای سم G2 و 8/8±97/0 برای سم B1 بود. بررسی آماری نشان داد اختلاف معنیداری بین مقادیر بازیابی و سموم اندازهگیری شده در نمونههای حقیقی با هر دو روش وجود نداشته و هر دو روش نیز از کارآیی کافی برخوردارند.

استخراج و ریزپوشانی کپسایسینویدهای اولئورزین فلفل قرمز
اولئورزین فلفل، عصاره روغنی غلیظ و یا تقریباً نیمهجامدی است که بهعنوان طعمدهنده و رنگدهنده در محصولات غذایی مختلف استفاده میشود. تندی فلفل قرمز به علت وجود گروهی از آلکالوئیدها موسوم به کپسایسینویدها است. در این پژوهش ابتدا استخراج اولئورزین فلفل قرمز با حلالهای اتانول، متانول، و 2- پروپانول انجام شد. سپس، ریزپوشانی کپسایسینویدهای موجود در اولئورزین فلفل قرمز با صمغ عربی و کنسانتره پروتئینی آب پنیر (WPC) بهعنوان مواد تشکیلدهنده دیواره کپسول به روش خشککن پاششی مورد بررسی قرار گرفت. تأثیر ترکیب دیواره (در غلظتهای 5، 10 و 15 درصد) و نیز نسبت اجزای سازنده ماده دیواره شامل صمغ عربی و WPC (با نسبت 100 درصد از هر یک و یا نسبت مساوی از هر دو) بر اندازه پودرهای حاصل از خشککردن امولسیون و راندمان کپسولهکردن کپسایسینوید ارزیابی شدند. نتایج آماری نشان داد که نوع حلال تأثیر معنیداری بر راندمان استخراج اولئورزین داشت درحالی که نسبت پودر به حلال بر روی راندمان استخراج اختلاف معنیداری نداشت. همچنین، در مرحله ریزپوشانی نوع و غلظت ماده دیواره تأثیر معنیداری بر متوسط قطر، توزیع اندازه و سطح مخصوص قطرات امولسیونهای حاصله داشت. بهطوری که صمغ عربی نسبت به WPC حفاظت بیشتری را در میزان نگهداری مواد مؤثره از خود نشان داد. علاوه بر این، دادهها حاکی از آن بود که با افزایش میزان غلظت ماده دیواره از 5 تا 15 درصد میزان ابقای مواد مؤثره افزایش یافت.

بررسی اثر آنتیاکسیدانی و ضدمیکروبی اسانس آویشن بر پنیر موزارلای نگهداری شده در دمای یخچال
هدف از این پژوهش، تولید پنیر موزارلا با طعم آویشن و بررسی اثر بازدارندگی اسانس Thymus vulgaris بر کپک، مخمر، اشرشیاکلی و کلیفرمها، برخی واکنشهای پروتئولیتیک و لیپولیتیک و افزایش زمان ماندگاری پنیر موزارلا در دمای یخچال میباشد. در این پژوهش آزمایشگاهی، اثر درصدهای مختلف اسانس آویشن (صفر، 0/001، 0/003 و 0/005) بر خصوصیات فیزیکوشیمیایی، میکروبی و آنتیاکسیدانی پنیر موزارلای نگهداری شده در دمای یخچال به مدت 45 روز مورد بررسی قرار گرفت. اسانس آویشن با غلظتهای 0/003 و 0/005 درصد، رشد کلیفرمها، اشرشیاکلی و کپک و مخمرها را بهطور معنیداری کاهش داد (0/05>P). همچنین غلظتهای 0/003 و 0/005 درصد اسانس، اندیس پراکسید را بهطور معنیداری در این فراورده کاهش داد (0/05>P) اما تأثیر معنیداری بر pH، اسیدیته، رطوبت نداشت (0/05>P). همچنین نمونههای حاوی 0/005 درصد اسانس، بهترین عطر و طعم و پذیرش کلی را داشتند. در نتیجه اسانس آویشن به عنوان یک نگهدارنده طبیعی، قادر به افزایش زمان ماندگاری پنیر موزارلا در دمای یخچال میباشد.

بررسی روشهای مختلف جداسازی و خواص فیزیکوشیمیایی فراکسیونهای با طول زنجیره مختلف اینولین حاصل از گیاه کاسنی
اینولین طبیعی استخراج شده از ریشههای تازه گیاه کاسنی متشکل از واحدهای فروکتوز با پیوندهای (1→2) βبا درجه پلیمریزاسیون مختلف میباشد. میزان اینولین، طول زنجیره و خواص عملکردی آن علاوه بر منشاء ژنتیکی، شرایط محیطی رشد و زمان برداشت گیاه، به روش استخراج و فرآیندهای پس از استخراج نیز بستگی دارد. اینولین با زنجیره کوتاه حلالیت و شیرینی بیشتری داشته و به عنوان جایگزین شکر استفاده میشود، حال آنکه اینولین زنجیره بلند دارای حلالیت کمتر، ویسکوزیته و پایداری حرارتی بیشتری بوده و به عنوان جایگزین چربی مورد استفاده قرار میگیرد. در این پژوهش جهت دستیابی به فراکسیون های اینولین با طول زنجیره مختلف، از ریشه گیاه کاسنی رقم ارکیس استفاده شد و اثر تیمارهای نسبت اتانل به ماده جامد (2:1 و 10:1)، کریستالیزاسیون و رسوب محلول آبی (20- درجه سانتیگراد) و اولترافیلتراسیون با قطر غشای 4 کیلودالتون بر بازده استخراج، درجه پلیمریزاسیون و خواص فیزیکوشیمیایی ترکیبات حاصل بررسی شد. نتایج نشان داد که افزایش غلظت اتانل باعث رسوب و راندمان بیشتر اینولین میشود. رسوب حاصل از نسبت 2:1 اتانل به ماده جامد در مقایسه با سایر روشهای استفاده شده دارای بیشترین درجه پلیمریزاسیون (66) و کمترین درصد قند احیاء بوده (1/2 درصد) و رسوب حاصل از فاز تراوه غشای اولترافیلتراسیون دارای بیشترین درصد قند احیاء (5/31 درصد) و کمترین درجه پلیمریزاسیون (16) بود.

روش نفوذ خشک مایکروویو و گرانش یکی از پیشرفتهترین روشهای استخراج به کمک مایکروویو است. هدف از این پژوهش، مقایسه روش استخراج نفوذ خشک مایکروویو و گرانش با روشهای تقطیر با آب به کمک مایکروویو و تقطیر با آب بر کیفیت اسانس رزماری بود. متغیرهایی چون بازدهی نهایی استخراج، زمان استخراج، ترکیبات شیمیایی، خاصیت آنتیاکسیدانی و میزان انرژی مصرفی مقایسه شدند. نتایج این پژوهش نشان داد زمان کل فرایند در روشهای استخراج اسانس به کمک نفوذ خشک مایکروویو و گرانش، تقطیر با آب به کمک مایکروویو و تقطیر با آب به ترتیب 5، 15 و 120 دقیقه و مصرف انرژی آنها به ترتیب 0/04، 0/13 و 1/00 کیلوواتساعت بود. روش استخراج اسانس بهکمک نفوذ خشک مایکروویو و گرانش مزایای مهمی از جمله، افزایش خاصیت آنتیاکسیدانی (10/3 درصد) و تولید اسانس با ارزشتر (آلفا پینن، کامفن و 1و-8-سینئول هر کدام به ترتیب 17/0، 2/1 و 2/8 درصد بیشتر) را نسبت به روش تقطیر با آب دارد. تجزیه و تحلیل کروماتوگرافی گازی/طیفسنج جرمی اسانسهای استخراج شده نشان داد که استفاده از امواج مایکروویو تأثیر مهمی روی ترکیب اسانسها ندارد. روش استخراج نفوذ خشک مایکروویو و گرانش نسبت به روشهای تقطیر با آب به کمک مایکروویو و تقطیر با آب نیاز به انرژی، هزینه و حلال کمتری داشته و به همین دلیل جزء فنآوریهای دوستدار محیط زیست شناخته میشود.

تولید پپتیدهای زیست فعال با فعالیت آنتیاکسیدانی از کنسانتره پروتئین آب پنیر
در تحقیق حاضر پروتئین هیدرولیز شده از کنسانتره پروتئین آب پنیر با بکارگیری آنزیم آلکالاز تولید شد. اثر متغیرهای دما (40، 45، 50 و 55 درجه سانتیگراد)، زمان (30، 60، 90، 120، 150، 180 و 210 دقیقه) و نسبت آنزیم به سوبسترا (30، 60 و 90 واحد آنسون بر کیلوگرم پروتئین) بر فعالیت آنتیاکسیدانی در قالب طرح کاملاً تصادفی بررسی گردید. فعالیت آنتیاکسیدانی پروتئین هیدرولیزشده آب پنیر توسط آزمونهای قدرت احیاءکنندگی و فعالیت شلاتهکنندگی یون آهن اندازهگیری شد. بالاترین فعالیت شلاتهکنندگی یون آهن در دما و فعالیت آنزیمی به ترتیب 50 درجه سانتیگراد و 60 واحد آنسون بر کیلوگرم و در زمان هیدرولیز 90 دقیقه به میزان 87/2 درصد بهدست آمد. بالاترین قدرت احیاءکنندگی پروتئینهای هیدرولیزشده نیز در دمای 40 درجه سانتیگراد، فعالیت آنزیمی90 واحد آنسون بر کیلوگرم و در زمان هیدرولیز 210 دقیقه و به میزان 0/435 حاصل شد که در مقایسه با اسیدآسکوربیک ppm 100، 57/08 درصد قدرت احیاءکنندگی از خود نشان داد. نتایج نشان میدهد که پپتیدهای آنتیاکسیدانی حاصل میتوانند بهعنوان آنتیاکسیدان طبیعی در فرمولاسیون غذایی و نیز بهعنوان ترکیب دارویی بهکار گرفته شوند.
