اثر هم‌زدن و هوادهی بر خصوصیات سینتیک رشد اسپیرولینا پلاتنسیس و تولید رنگ‌دانه‌های طبیعی در فتوبیوراکتور همزن‌دار

چکیده

اسپیرولینا پلاتنسیس یک سیانوباکتریوم رشته‌ای فتوسنتزکنندۀ پلانکتونی است که حاوی مولکول‌های فعال‌زیستی و منبع غنی از رنگ‌دانه‌هایی مانند فیکوسیانین است. در این پژوهش تأثیر دو عامل مهم هم‌زدن (20 و 50 دور در دقیقه) و هوادهی (با هوادهی و بدون هوادهی) بر کشت ریزجلبک اسپیرولینا پلاتنسیس و تولید رنگ‌دانه‌های طبیعی (فیکوسیانین، آلوفیکوسیانین، کلروفیل و کاروتنوئید) در شرایط دمای 28 درجۀ سانتی‌گراد، 9=pH، در کشت غوطه‌وری در فرمانتور همزن‌دار موردمطالعه قرار گرفت. یافته‌ها نشان داد هوادهی در تیمارهای با سرعت هم‌زدن 20 دور در دقیقه باعث افزایش معنی‌دار غلظت رنگ‌دانه‌ها (فیکوسیانین، آلوفیکوسیانین، کلروفیل و کاروتنوئید) و زیست‌توده شد، درحالی‌که هوادهی در تیمارهای با سرعت 50 دور در دقیقه باعث مهار تولید این ترکیبات گردید. بیشترین غلظت زیست‌توده، فیکوسیانین، آلوفیکوسیانین، کلروفیل و کاروتنوئید به‌ترتیب 1/39 گرم در لیتر، 136/5، 38، 8/62 و 3/05 میلی‌گرم در لیتر مربوط به تیمار 20 دور در دقیقه با هوادهی بود. باتوجه‌به نتایج به‌دست‌آمده، هوادهی محیط کشت به‌صورت معنی‌داری غلظت آلوفیکوسیانین را افزایش داد (0/05≤P). در شرایط بدون هوادهی، افزایش سرعت هم‌زدن، غلظت زیست‌توده را افزایش داد. در سرعت هم‌زدن 20 دور در دقیقه همراه با هوادهی بیشترین غلظت زیست‌توده، فیکوسیانین، آلوفیکوسیانین، کلروفیل و کاروتنوئید به‌دست‌آمد.
